Testing for Sperm DNA Fragmentation

When general sperm parameters such as – motility, concentration, morphology have been within the normal range, sperm may still be the cause of unsuccessful results after artificial insemination procedure. Sperm DNA fragmentation is expressed as a rupture of DNA strand in both or just one DNA helix. If the rupture affects genes which are vital for embryo and implantation development then during some stage the embryo may not survive.

Possible causes of DNA fragmentation:

  • Oxidative stress
  • Infections
  • Drug use 
  • Smoking
  • Environmental pollution (heavy metal, air pollution) 
  • Age 
  • Nutrition 
  • Elevated testicular temperature etc.

DNA fragmentation test is recommended for patients with:

  • Unexplained Infertility 
  • Cessation of development of embryo 
  • Poor blastocyst development 
  • Multiple unsuccessful treatment of cycles 
  • Pregnancy interruption in short period of time (miscarriages)
  • Varicocele, etc.

In some cases, DNA fragmentation of sperm may be reduced by changing your way of life and habits. In addition, eating antioxidants contained in food will prevent the formation of free-radicals and may prevent diseases caused by DNA fragmentation.

Sperm DNA fragmentation test is a laboratory diagnostic tool. It is based on the processing of sperm sample with various reagents and visualisation in the fluorescence microscope.
